Abstract

Milk thistle (Silybum marianum) is a significant medicinal plant used to treat various diseases, particularly liver diseases and cancer. The primary bioactive compound in this plant is silymarin, which has potent medicinal properties. An effective strategy to enhance silymarin production is the use of elicitors, which stimulate the expression of genes involved in its biosynthetic pathway. Multi-walled carbon nanotubes (MWCNT) are one such elicitor known in plant sciences for their ability to promote growth, improve stress resistance, and increase the production of secondary metabolites. In this study, we investigated the impact of varying MWCNT concentrations (0, 150, 300, and 600 ppm) and spraying durations (24 and 48 h) on the expression of thirteen genes associated with secondary metabolite production in S. marianum. Our results indicated that a 48-hour foliar spray with 600 ppm MWCNT significantly enhanced the expression of several genes: PAL (9.42-fold), CHI (4.99-fold), CHS1 (14.39-fold), CHS2 (15.57-fold), CHS3 (6.85-fold), CHS11 (12.03-fold), SmTIP41 (6.10-fold), SmMON1 (6.10-fold), and SmNDHF (3.90-fold). Additionally, a 24-hour spraying interval at the same 600 ppm concentration resulted in increased expression of SmUBC (6.18-fold), SmRNASE3 (12.88-fold), and SmPRX1 (13.95-fold). The highest induction of CHS33 gene expression was observed with the 600 ppm treatment, regardless of the duration of spraying. Furthermore, Pearson's correlation analysis revealed a positive and significant association among the upregulated expressions of the studied genes. In conclusion, our findings suggest that a foliar application of 600 ppm MWCNT for 48 h most effectively enhances the expression of genes associated with the silymarin synthesis pathway.
