Abstract

Salmonella Typhimurium remains a leading cause of foodborne gastroenteritis globally. However, verifying bacterial inactivation during food processing is hampered by the laborious nature of culture methods and the inability of current detection tools to discriminate viable from inactivated cells. This study presents a conformation-sensitive nanobody-based immunoassay that selectively recognizes heat-inactivated Salmonella Typhimurium. Nanobodies were generated via alpaca immunization and phage display, and two enzyme-linked immunosorbent assay (ELISA) were established: a direct ELISA and a sandwich ELISA enhanced by streptavidin-PolyHRP.
