Abstract

As a three-dimensional miniature organ model, organoids can simulate the structural and functional heterogeneity of the tissues, especially demonstrating unique advantages in disease pathogenesis and personalized drug screening. Exosomes are key carriers of intercellular communication, carrying small biological molecules (proteins and nucleic acids), and exhibit the significant potential in disease diagnosis, signal transduction, and targeted therapy. However, both of them have certain limitations in terms of applications. The synergistic applications of organoids and exosomes can achieve complementary functions and information carriers between them, which may become a potential strategy to promote the elucidation of disease mechanisms, drug screening, and the clinical translation. In this review, we summarize the cultivation and isolation methods of organoids and exosomes, and introduce the synergistic applications of organoid and exosomes in disease modeling, mechanism research, diagnostic biomarker discovery, and treatment strategy development in different types of diseases. This review will establish a foundation for theoretical basis and technical reference for precision medicine and regenerative medicine.
