Immune-Mediated Gut-Lung Axis in IPF: Bidirectional Mendelian Randomization and Mediation Analysis of Microbiota, Inflammatory Proteins, and Metabolites.
Article in Endocrine, metabolic & immune disorders drug targets, 2026. The graph could read no effect estimate from its abstract, so it casts no vote on the map. Not yet cited in PubMed.
What it found
Each row is one number read from the abstract, on the scale the paper reported it, with its interval. Left of the dashed line favours the treatment, right favours the comparator. Under each row is the sentence it came from. New to these charts? A ten-minute tutorial.
The abstract states no effect estimate the extractor could read, or names no intervention and outcome on the map, so this paper lights no cell and moves no belief. It is still indexed, cited and linked below.
The trial behind it
Trials whose registry record cites this paper, or whose number appears in the abstract. A trial that started after this paper was published is citing it as background, not reporting it.
Neither the registry nor the abstract names a trial number. If this is a trial report, that itself is worth knowing.

Abstract
introductionImmune dysregulation plays a critical role in idiopathic pulmonary fibrosis (IPF), but the mechanisms underlying this dysregulation remain unclear.
methodsWe performed bidirectional Mendelian randomization (MR) and mediation analysis using genome-wide association studies (GWAS) summary data from FinnGen (731 immune cell phenotypes, 473 gut microbiota, 91 inflammatory proteins, 179 plasma lipidome, 1,400 metabolites, and IPF cases). Sensitivity analyses ensured robustness.
resultsNine immune cell phenotypes, nine gut microbiota taxa, three inflammatory proteins, one lipid species, and 29 metabolites showed significant causal associations with IPF. Regulatory T cells (Tregs) showed a dual role: protective subsets reduced IPF risk, while activated CD4+ Tregs increased susceptibility. Mediation analysis revealed that the effect of activated CD4+ Tregs was partially mediated by DISCUSSION: These findings enhance our understanding of IPF pathogenesis, suggesting that modulating key metabolic pathways and immune regulatory nodes, such as Tregs, could represent promising therapeutic approaches. However, additional experimental studies are needed to validate these potential causal relationships.
conclusionThis MR study systematically maps multi-omics interactions in IPF, identifying novel mediators and therapeutic targets. Validating these mechanisms in experimental models could advance precision immunomodulatory strategies for IPF.
