1 · What the graph read from it

What it found

Each row is one number read from the abstract, on the scale the paper reported it, with its interval. Left of the dashed line favours the treatment, right favours the comparator. Under each row is the sentence it came from. New to these charts? A ten-minute tutorial.

The abstract states no effect estimate the extractor could read, or names no intervention and outcome on the map, so this paper lights no cell and moves no belief. It is still indexed, cited and linked below.

2 · The registry

The trial behind it

Trials whose registry record cites this paper, or whose number appears in the abstract. A trial that started after this paper was published is citing it as background, not reporting it.

Neither the registry nor the abstract names a trial number. If this is a trial report, that itself is worth knowing.

Abstract

A Disintegrin and Metalloprotease with ThromboSpondin type 1 repeats, 13 (ADAMTS13) is an anti-thrombotic metalloenzyme that cleaves ultra-large von Willebrand factor, preventing platelet accumulation at sites of vascular injury. Deficiency of plasma ADAMTS13 activity leads to thrombotic thrombocytopenic purpura and is a risk factor for the development of other inflammatory thrombotic disorders. However, it is not known if ADAMTS13 has other biological functions. The present study aims to determine the effects of ADAMTS13 on agonist-induced platelet activation and aggregation and its underlying molecular mechanisms.A light transmission assay was used to determine platelet aggregation; flow cytometry was used to determine platelet activation, phosphatidylserine (PS) exposure, and AKT phosphorylation.A full-length recombinant ADAMTS13 but not its truncated form (MDTCS or TSP2-CUB) enhanced a fibrillar collagen-induced aggregation of isolated human and murine platelets. This enhancing effect was ADAMTS13 concentration- and conformation-dependent but did not depend on its proteolytic activity. Interestingly, recombinant ADAMTS13 did not enhance AYPGKF, an agonist of protease-activated receptor 4, or 2MeSADP-induced murine platelet aggregation. Flow cytometry demonstrated that recombinant ADAMTS13 plus a fibrillar collagen resulted in significantly more activation and PS exposure of murine platelets than the collagen did alone, likely via an increase in AKT phosphorylation. Our results demonstrate a novel but paradoxical role of recombinant ADAMTS13, possibly promoting hemostasis while inhibiting thrombosis at the same time. The findings provide novel insight into the excellent therapeutic efficacy of recombinant ADAMTS13 for thrombosis but without an increased risk of bleeding.
